马来簕竹的组织培养与快速繁殖

摘 要:

马来簕竹是以修长叶形及优良株形作为观赏性状的重要观赏竹之一。本文以马来簕竹带芽茎段为外植体, 研究植物生长调节剂对其快速繁殖及生根的影响。结果表明: 茎段腋芽萌发的最佳培养基为MS+2.0 mg•L-1 6-BA+0.5 mg•L-1 NAA+0.001 mg•L-1 TDZ+30.0 g•L-1蔗糖+6.5 g•L-1琼脂, 萌芽率达72.22%; 最佳继代增殖培养基为MS+3.0 mg•L-1 6-BA+0.5 mg•L-1 NAA+0.01 mg•L-1 TDZ+30.0 g•L-1蔗糖+6.5 g•L-1琼脂, 增殖系数可达3.63; 最佳生根培养基为MS+1.0 mg•L-1 NAA+0.5 mg•L-1 IBA+20.0 g•L-1蔗糖+7.0 g•L-1琼脂, 生根率为100%; 待根生长至5~10 cm时, 将生根组培苗炼苗后盆栽, 30 d后成活率 达95.60%。

Abstract:

Bambusa glaucophylla is one of the most beautiful ornamental bamboo species, which is famous for its slender leaves and excellent shape. The rapid propagation system and rooting in vitro were studied using culm internodes with axillary buds as the explants. The media with different plant growth substances for axillary buds induction, subculture and rooting culture were selected. The results show that the optimal medium for germination of culm axillary bud was MS+2.0 mg•L-1 6-BA+0.5 mg•L-1 NAA+0.001 mg•L-1 TDZ+30.0 g•L-1 sugar+6.5 g•L-1 agar, with a 72.22% germination rate of axillary buds; the suitable medium for multiplication was MS+3.0 mg•L-1 6-BA+0.5 mg•L-1 NAA+0.01 mg•L-1 TDZ+30.0 g•L-1 sugar+6.5 g•L-1 agar, and the multiplication coefficient could reach up to 3.63; the optimized medium for rooting was MS+1.0 mg•L-1 NAA+0.5 mg•L-1 IBA+20.0 g•L-1 sugar+7.0 g•L-1 agar, with a 100% rooting rate. The young seedlings with roots of 5–10 cm were transplanted into greenhouse, the survival rate were 95.60% 30 days later.
